Accurate recognition of colorectal cancer with semi-supervised deep learning on pathological images. Nature Communications, 2021; 12 (1) DOI: 10.1038/s41467-021-26643-8

The study, which was conducted by researchers from Tulane, Central South University in China, the University of Oklahoma Health Sciences Center, Temple University, and Florida State University, was designed to test whether AI could be a tool to help pathologists keep pace with the rising demand for their services.

Pathologists evaluate and label thousands of histopathology images on a regular basis to tell whether someone has cancer. But their average workload has increased significantly and can sometimes cause unintended misdiagnoses due to fatigue.

“Even though a lot of their work is repetitive, most pathologists are extremely busy because there’s a huge demand for what they do but there’s a global shortage of qualified pathologists, especially in many developing countries” said Dr. Hong-Wen Deng, professor and director of the Tulane Center of Biomedical Informatics and Genomics at Tulane University School of Medicine. “This study is revolutionary because we successfully leveraged artificial intelligence to identify and diagnose colorectal cancer in a cost-effective way, which could ultimately reduce the workload of pathologists.”

To conduct the study, Deng and his team collected over 13,000 images of colorectal cancer from 8,803 subjects and 13 independent cancer centers in China, Germany and the United States. Using the images, which were randomly selected by technicians, they built a machine assisted pathological recognition program that allows a computer to recognize images that show colorectal cancer, one of the most common causes of cancer related deaths in Europe and America.

“The challenges of this study stemmed from complex large image sizes, complex shapes, textures, and histological changes in nuclear staining,” Deng said. “But ultimately the study revealed that when we used AI to diagnose colorectal cancer, the performance is shown comparable to and even better in many cases than real pathologists.”

The area under the receiver operating characteristic (ROC) curve or AUC is the performance measurement tool that Deng and his team used to determine the success of the study. After comparing the computer’s results with the work of highly experienced pathologists who interpreted data manually, the study found that the average pathologist scored at .969 for accurately identifying colorectal cancer manually. The average score for the machine-assisted AI computer program was .98, which is comparable if not more accurate.

Using artificial intelligence to identify cancer is an emerging technology and hasn’t yet been widely accepted. Deng’s hope is that the study will lead to more pathologists using prescreening technology in the future to make quicker diagnoses.

“It’s still in the research phase and we haven’t commercialized it yet because we need to make it more user friendly and test and implement in more clinical settings. But as we develop it further, hopefully it can also be used for different types of cancer in the future. Using AI to diagnose cancer can expedite the whole process and will save a lot of time for both patients and clinicians.”

Terahertz sources emit brief light pulses oscillating at ‘trillion of times per second’. At this scale, they are too fast to be handled by standard electronics, and, until recently, too slow to be handled by optical technologies. This has great significance for the evolution of ultra-fast communication devices above the 300GHz limit — such as that required for 6G mobile phone technology — something that is still fundamentally beyond the limit of current electronics.

Researchers in the Emergent Photonics (EPic) Lab at Sussex, led by the Director of the Emergent Photonics (EPic) Lab Professor Marco Peccianti, are leaders in surface terahertz emission technology having achieved the brightest and thinnest surface semiconductor sources demonstrated so far. The emission region of their new development, a semiconductor source of terahertz, is 10 times thinner than previously achieved, with comparable or even better performances.

The thin layers can be placed on top of existing objects and devices, meaning they are able to place a terahertz source in places that would have been inconceivable otherwise, including everyday object such as a teapot or even a work of art — opening up huge potential for anti-counterfeiting and ‘the internet of things’ — as well as previously incompatible electronics, such as a next generation mobile phone.

Dr Juan S. Totero Gongora, Leverhulme Early Career Fellow at the University of Sussex, said:

“From a physics perspective, our results provide a long-sought answer that dates back to the first demonstration of terahertz sources based on two-colour lasers.

“Semiconductors are widely used in electronic technologies but have remained mostly out of reach for this type of terahertz generation mechanism. Our findings therefore open up a wide range of exciting opportunities for terahertz technologies.”

Dr Luke Peters, Research Fellow of the European Research Council project TIMING at the University of Sussex, said:

“The idea of placing terahertz sources in inaccessible places has great scientific appeal but in practice is very challenging. Terahertz radiation can have a superlative role in material science, life science and security. Nevertheless, it is still alien to most of the existing technology, including devices that talk to everyday objects as part of the rapidly expanding ‘internet of things’.

” This result is a milestone in our route to bring terahertz functions closer to our everyday lives.”

Lying between microwaves and infrared in the electromagnetic spectrum, terahertz waves are a form of radiation highly sought in research and industry. They have a natural ability to reveal the material composition of an object by easily penetrating common materials like paper, clothes and plastic in the same way X-rays do, but without being harmful.

Terahertz imaging makes it possible to ‘see’ the molecular composition of objects and distinguish between different materials. Previous developments from Prof Peccianti’s team showcased the potential applications of terahertz cameras, which could be transformative in airport security, and medical scanners — such as those used to detect skin cancers.

One of the biggest challenges faced by scientists working in terahertz technology is that what is commonly accepted as an ‘intense terahertz source’ is faint and bulky when compared with, for example, a light bulb. In many cases, the need for very exotic materials, such as nonlinear crystals, makes them unwieldy and expensive. This requirement poses logistical challenges for integration with other technologies, such as sensors and ultrafast communications.

The Sussex team have overcome these limitations by developing terahertz sources from extremely thin materials (about 25 atomic layers). By illuminating an electronic-grade semiconductor with two different types of lasers light, each oscillating at different frequency or colour, they were able to elicit the emission of short bursts of Terahertz radiation.

This scientific breakthrough has been long-sought by scientists working in the field since the first demonstration of terahertz sources based on two-colour lasers in the early 2000s. Two-colour terahertz sources based on special mixtures of gas, such as nitrogen, argon or krypton, are among the best performing sources available today. Semiconductors, widely used in electronic technologies, have remained mostly out of reach for this type of terahertz generation mechanism.

In the fight to eradicate poverty around the world and achieve decent living standards (DLS), having sufficient energy is a key requirement. Despite international commitments such as the UN Sustainable Development Goals, in many areas progress on achieving DLS worldwide has been slow. There are also fears that improving energy access could lead to higher carbon dioxide emissions, which would interfere with goals to alleviate climate change.

In a new study published in the journal Environmental Research Letters, IIASA researchers used a multidimensional approach to poverty to conduct a comprehensive global study on DLS. The researchers identified gaps in DLS by region and estimated how much energy is needed to fill them. They also assessed whether providing everyone with a decent life is compatible with climate goals

Studies on poverty often use an income-based definition for defining poverty thresholds ($1.90/day or $5.50/day), which obscures that there are other factors contributing to human wellbeing more directly. In contrast, DLS represent a set of material prerequisites to provide the services needed for wellbeing, such as having adequate shelter, nutrition, clean water, sanitation, cooking stoves and refrigeration, and being able to connect physically and socially via transportation and communication technologies. Crucially, this allows for calculation of the resources needed to provide these basic services.

The largest gaps in DLS were found in sub-Saharan Africa, where more than 60% of the population are lacking in at least half of the DLS indicators. The researchers also identified high DLS deprivation in indicators such as sanitation and water access, access to clean cooking, and thermal comfort in South and Pacific Asia, and more moderate gaps in other regions. One of the most striking findings of the study was that the number of people deprived of basic needs according to DLS generally far exceeds the number of people in extreme poverty, meaning that current poverty thresholds are often inconsistent with a decent life.

When looking at which components of DLS require the most investment in energy, the researchers identified shelter and transport as having the largest share.

“The majority of the global population does not currently have decent levels of motorized transport. An important policy lesson for national governments is the large impact of investing in public transit to reduce the use of passenger vehicles, which generally have much higher energy use per person,” says Jarmo Kikstra, lead author of the study and a researcher in the IIASA Energy, Climate, and Environment Program.

The upfront energy required globally to build new houses, roads, and other materials to enable DLS provision for all from 2015 to 2040 is about 12 exajoules per year. This is only a fraction of current total final energy use, which exceeds 400 exajoules per year. The increase in annual energy for operating this increase in services, including maintenance costs, is more substantial, eventually increasing by about 68 exajoules. For some countries, reaching this goal would require robust changes in development, which will be challenging, especially in the Global South.

“For most countries, especially many poor countries in Africa, unprecedented growth in energy use as well as more equitably distributed growth are essential to achieving DLS before mid-century,” Kikstra adds. “Therefore, the biggest challenge for policymakers will be to achieve an equitable distribution of energy access worldwide, which is currently still out of reach.”

According to the study, the amount of energy needed for decent living worldwide is less than half of the total final energy demand projected under most future pathways that keep temperature rise below 1.5° C. This indicates that achieving DLS for all does not have to interfere with climate goals. While this ratio changes in different climate mitigation scenarios and by region, the energy needs for DLS always remain well below the projected energy demands on the level of larger global regions.

“To achieve decent living conditions worldwide, it seems that we do not have to limit energy access to basic services as there is a surplus of total energy. What is perhaps unexpected is that even under very ambitious poverty eradication and climate mitigation scenarios, there is quite a lot of energy still available for affluence,” says study author Alessio Mastrucci.

“Our results support the view that on a global scale, energy for eradicating poverty does not pose a threat for mitigating climate change. However, to provide everyone with a decent life, energy redistribution across the world and unprecedented final energy growth in many poor countries is required,” concludes study author, Jihoon Min.

“This study points the way to a new paradigm that is desperately needed in psychiatry,” said Andrew Krystal, PhD, professor of psychiatry and member of the UCSF Weill Institute for Neurosciences. “We’ve developed a precision-medicine approach that has successfully managed our patient’s treatment-resistant depression by identifying and modulating the circuit in her brain that’s uniquely associated with her symptoms.”

Previous clinical trials have shown limited success for treating depression with traditional deep brain stimulation (DBS), in part because most devices can only deliver constant electrical stimulation, usually only in one area of the brain. A major challenge for the field is that depression may involve different brain areas in different people.

What made this proof-of-principle trial successful was the discovery of a neural biomarker — a specific pattern of brain activity that indicates the onset of symptoms — and the team’s ability to customize a new DBS device to respond only when it recognizes that pattern. The device then stimulates a different area of the brain circuit, creating on-demand, immediate therapy that is unique to both the patient’s brain and the neural circuit causing her illness.

This customized approach alleviated the patient’s depression symptoms almost immediately, Krystal said, in contrast to the four- to eight-week delay of standard treatment models and has lasted over the 15 months she has had the implanted device. For patients with long-term, treatment-resistant depression, that result could be transformative.

“I was at the end of the line,” said the patient, who asked to be known by her first name, Sarah. “I was severely depressed. I could not see myself continuing if this was all I’d be able to do, if I could never move beyond this. It was not a life worth living.”

Applying Proven Advances in Neuroscience to Mental Health

The path to this project at UCSF began with a large, multicenter effort sponsored under President Obama’s BRAIN (Brain Research through Advancing Innovative Neurotechnologies) Initiative in 2014.

Through that initiative, UCSF neurosurgeon Edward Chang, MD, and colleagues conducted studies to understand depression and anxiety in patients undergoing surgical treatment for epilepsy, for whom mood disorders are also common. The research teamdiscovered patterns of electrical brain activity that correlated with mood states and identified new brain regions that could be stimulated to relieve depressed mood.

With results from the previous research as a guide, Chang, Krystal, and first author Katherine Scangos, MD, PhD, all members of the Weill Institute, developed a strategy relying on two steps that had never been used in psychiatric research: mapping a patient’s depression circuit and characterizing her neural biomarker.

“This new study puts nearly all the critical findings of our previous research together into one complete treatment aimed at alleviating depression,” said Chang, who is co-senior author with Krystal on the paper and the Joan and Sanford Weill Chair of Neurological Surgery.

The team evaluated the new approach in June 2020 under an FDA investigational device exemption, when Chang implanted a responsive neurostimulation device that he has successfully used in treating epilepsy.

“We were able to deliver this customized treatment to a patient with depression, and it alleviated her symptoms,” said Scangos. “We haven’t been able to do this the kind of personalized therapy previously in psychiatry.”

To personalize the therapy, Chang put one of the device’s electrode leads in the brain area where the team had found the biomarker and the other lead in the region of Sarah’s depression circuit where stimulation best relieved her mood symptoms. The first lead constantly monitored activity; when it detected the biomarker, the device signaled the other lead to deliver a tiny (1mA) dose of electricity for 6 seconds, which caused the neural activity to change.

“The effectiveness of this therapy showed that not only did we identify the correct brain circuit and biomarker, but we were able to replicate it at an entirely different, later phase in the trial using the implanted device,” said Scangos. “This success in itself is an incredible advancement in our knowledge of the brain function that underlies mental illness.”

Translating Neural Circuits into New Insights

For Sarah, the past year has offered an opportunity for real progress after years of failed therapies.

“In the early few months, the lessening of the depression was so abrupt, and I wasn’t sure if it would last,” she said. “But it has lasted. And I’ve come to realize that the device really augments the therapy and self-care I’ve learned while being a patient here at UCSF.”

The combination has given her perspective on emotional triggers and irrational thoughts on which she used to obsess. “Now,” she said, “those thoughts still come up, but it’s just…poof…the cycle stops.”

While the approach appears promising, the team cautions that this is just the first patient in the first trial.

“There’s still a lot of work to do,” said Scangos, who has enrolled two other patients in the trial and hopes to add nine more. “We need to look at how these circuits vary across patients and repeat this work multiple times. And we need to see whether an individual’s biomarker or brain circuit changes over time as the treatment continues.”

FDA approval for this treatment is still far down the road, but the study points toward new paths for treating severe depression. Krystal said that understanding the brain circuits underlying depression is likely to guide future non-invasive treatments that can modulate those circuits.

Added Scangos, “The idea that we can treat symptoms in the moment, as they arise, is a whole new way of addressing the most difficult-to-treat cases of depression.”

Samuel L. Jackson shares a new photo from Secret Invasion that teases young Nick Fury scenes, possibly set before the events of Captain Marvel.

Samuel L. Jackson teases he’s filming some young Nick Fury scenes for Secret Invasion. The actor has been part of the Marvel Cinematic Universe since the beginning, as Nick Fury was part of Iron Man‘s post-credits scene. Samuel L. Jackson has played a prominent role in the shared universe ever since, making 10 appearances overall. However, Secret Invasion is the first MCU project that will put the former SHIELD director in the lead role.

Marvel Studios announced plans for Secret Invasion in late 2020 as a new MCU series for Disney+. The limited series is a loose adaptation of the comic crossover event of the same name, which revealed the shape-shifting aliens known as Skrulls secretly invaded Earth for decades and replaced some of Marvel’s biggest characters. Secret Invasion so far doesn’t have any major MCU heroes confirmed to star, as Nick Fury, Talos (Ben Mendelsohn), and Maria Hill (Cobie Smulders) are the only familiar faces officially returning. Exact plot details are still under wraps too, although viewers can expect a thrilling story involving Fury and the Skrulls.

The cast and crew of Secret Invasion have been busy working on the show for the last few months. Jackson is known for sharing updates on his various projects on social media, which is how he’s now delivered a tease for young Nick Fury scenes. The actor’s latest Instagram post shows him preparing to film scenes for Secret Invasion and sporting Fury’s usual goatee, not the long grey beard previously seen. This seems to be flashback scenes as Jackson doesn’t have Fury’s usual eyepatch or scars, and his caption references getting back into his “Old School Fury” groove. Check out the image below:

Secret Invasion wouldn’t be the first MCU property to feature a young Nick Fury in some fashion if that is indeed what Jackson is teasing. There is always the chance he took this photo before the scar makeup was applied or he had to put the eyepatch back on. However, the caption makes it seem as though Jackson wants fans to know Secret Invasion will feature a younger Nick Fury at times. Marvel Studios pulled this off quite well with Captain Marvel, which de-aged Jackson by several decades to play Fury when he was still just a SHIELD agent. Secret Invasion could do the same and flesh out some of Fury’s past in the process.

That said, Jackson’s tease that a younger Fury scene will happen without his eyepatch or scars is quite intriguing. The presence of Fury’s goatee means that the flashback can’t take place during Captain Marvel, as Fury didn’t have any facial hair during the movie’s events. Meanwhile, the mention of no eyepatch or scars means going back before Captain Marvel‘s events. This would be before Fury ever had any contact with the Skrulls or superheroes, so perhaps Secret Invasion plans to show more of Nick Fury’s origin.

Live.A$AP”.

Kendall y Kylie Jenner

Indie genre specialist XYZ Films is getting into the distribution game, launching a U.S. operation and bringing on board former Drafthouse Films and Neon executive James Emanuel Shapiro.

The domestic operation — which Shaprio will run as executive vp of U.S. distribution — will launch with Something in the Dirt, the latest feature from directing duo Justin Benson and Aaron Moorhead, which is billed as a return to their DYI indie origins seen in features like Resolution (2012) and Spring (2014). Produced by their longtime producer David Lawson under their Rustic Films production company banner, Something in the Dirt is currently in post. The feature marks the fifth collaboration with XYZ. More recently, Benson and Moorhead have seen mainstream success with sci-fi thriller Synchronic (2019) starring Anthony Mackie and Jamie Dornan, which hit #1 on its Netflix debut. The duo is currently shooting Marvel series Moon Knight for Disney+.

Shapiro, who said coming on board XYZ Film was “a dream come true” called Something in the Dirt the perfect film to launch the new division.

“Justin, Aaron, and Dave were incredible to work with during the Drafthouse Films release of Spring and I’ve loved each film they’ve done since,” he said. “Something in the Dirt is smart, inventive, and very entertaining. I hope we can distribute every film they do going forward.”

XYZ said their U.S. company will focus on the kind of auteur-driven genre films the group is known for. Since Nate Bolotin, Nick Spicer, and Aram Tertzakian founded XYZ in 2008, the company has delivered such cult hits as Gareth Evans’ The Raid, Panos Cosmatos’ Nicolas Cage revenge thriller Mandy, and Joe Penna’s sci-fi feature Stowaway starring Anna Kendrick and Toni Collette, another top hit on Netflix.

“We specialize in a certain kind of film, and we know that audience inside out,” said XYZ CEO Spicer, and partner Bolotin. “We now have the ability to be a true studio to our filmmakers, all the way through release. James has been distributing elevated, indie genre films for twenty years and knows how to reach that audience better than anyone.”

A 20-year veteran of the indie film scene, Shapiro has worked on the Oscar awards campaigns on such films as Bullhead (2011), The Act of Killing (2012), and I, Tonya (2017). The former COO of Drafthouse Films and Neon, he’s also familiar with the genre festival circuit, having worked with both the Fantastic Fest and Beyond Fest.

Shapiro will report to XYZ Films’ CEO Spicer and XYZ Films’ Partner Bolotin and will work alongside president of international sales and distribution Tatyana Joffe on global releases from XYZ’s slate.

“With traditional distribution models rapidly shifting, we need to quickly adapt to the demands of our international partners and their audience,” said Joffe. “Having control of the windowing, and working collectively with our key international partners to create marketing campaigns will greatly contribute to our global release strategy in capturing broader audience, which will ultimately ensure better performance worldwide.”

A production, sales and — most recently — management group, XYZ Films greatly boosted its financing capabilities in 2019 when it raised $100 million together with Finland-based venture capital firm IPR.VC to fund a slate of independent genre films.

The company said the U.S. division would be a boutique operation and wouldn’t release every film that XYZ finances or produces.

XYZ announced its U.S. distribution company as Cannes Virtual Film Market kicked off Monday. Among the new titles on XYZ’s Cannes slate are the Cambodian sci-fi feature Karmalink, which XYZ is selling for North America, with Scandinavian outfit LevelK handling for the rest of the world.
